HDM2 ERKs PCNA
In this issue, a study by Groehler and Lannigan (2010. J. Cell Biol. doi:10.1083/jcb.201002124) sheds light on the regulation of proliferating cell nuclear antigen (PCNA) turnover and how it is counteracted by the small chromatin-bound kinase ERK8 (extracellular signal-regulated kinase 8). Importantly, inactivation of ERK8 results in genome instability and is associated with cell transformation.
متن کاملThe archaeal PCNA proteins.
PCNA (proliferating-cell nuclear antigen) is a ring-shaped protein that encircles duplex DNA and plays an essential role in many DNA metabolic processes. The PCNA protein interacts with a large number of cellular factors and modulates their enzymatic activities. In the present paper, we summarize the structures, functions and interactions of the archaeal PCNA proteins.
متن کاملEmbryonic PCNA: a missing link?
PCNA, the processivity factor of the eukaryotic replicative DNA polymerase [1], is the functional homolpgue of the [3 subunit of Escherichia coli DNA polymerase III holo-enzyme [2,3]. Biochemical studies have shown that the E. coli 3 subunit acts as a sliding clamp on duplex DNA; it has a ring shape that encircles DNA and it binds directly to Pol III, tethering it to DNA to achieve highly proce...
متن کاملPCNA and DNA replication ( I )
Replication of the entire genome during S-phase is one of the most important cell cycle events and requires the coordinated activity of a multitude of different enzymes. Proliferating cell nuclear antigen (PCNA) interacts directly and indirectly with most of these proteins and thereby plays a leading part in the coordination of the replication process. PCNA is the processivity factor for the re...
متن کاملInhibition of DNA replication by an anti-PCNA aptamer/PCNA complex
Proliferating cell nuclear antigen (PCNA) is a multifunctional protein present in the nuclei of eukaryotic cells that plays an important role as a component of the DNA replication machinery, as well as DNA repair systems. PCNA was recently proposed as a potential non-oncogenic target for anti-cancer therapy. In this study, using the Systematic Evolution of Ligands by EXponential enrichment (SEL...
